The prefecture is divided into 23 municipalities and 3 communities. The capital and largest city of the prefecture is Trikala. It is a commercial, industrial, administrative and ecclesiastical center. The next most populous city is Kalambaka.

On May Day, the custom of Klidonas in Koziakas is worth seeing, where all the girls and boys of the village participate. The Sarakatsanai festival in the meadows of Pertouli at the end of June every year is also noteworthy. Also, in all the villages of the plains and the mountains, the famous traditional festivals are organized, where the inhabitants feast with folk songs and dances.
